Job Description

The candidate will focus on Dyskeratosis congenita (DC), a genetic syndrome characterized by short telomeres, and will investigate the underlying mechanisms of bone marrow failure and immune dysfunctions. The project involves creating a DC human model using CRISPR-Cas9 to study the effects of telomere shortening on hematopoietic stem and progenitor cells (HSPC). The research will include:

  • Imaging and flow cytometry to examine DNA damage response (DDR).
  • Single-cell RNA sequencing and histone mark analysis for transcriptional and epigenetic changes.
  • Luminex assays for proinflammatory phenotype assessment.
  • Functional experiments with colony-forming assays and long-term hematopoietic reconstitution.
  • Validation with patient-derived HSPCs in collaboration with Gaslini Hospital, Genoa.
