Summary
The Institute for Integrative Biology of the Cell (I2BC) is seeking a highly motivated Postdoctoral Researcher to join the ERC Synergy project 3Stops2go. This project focuses on engineering alternative decoding mechanisms for therapeutic purposes, specifically identifying novel suppressor tRNAs and elucidating the molecular mechanism of readthrough inducers. The successful candidate will contribute to designing quantitative reporters, implementing functional screens, analyzing high-throughput data, and interpreting results.

Skills
- PhD in Molecular Biology, Biochemistry, or a related field.
- Solid expertise in RNA and translation.
- Proficiency in cloning, human cell culture, flow cytometry, and NGS data analysis.
- Experience with lentiviral vectors, functional screens, or ribosome profiling is a significant asset.

Job Description
Missions
The postdoctoral researcher will participate in the ERC Synergy project 3Stops2go, which aims to engineer alternative decoding for therapeutic applications. The project at I2BC has two main objectives:
- To design, implement, and analyze a large-scale screen in human cells to identify novel suppressor tRNAs capable of efficiently and selectively reading through premature termination codons (PTCs).
- To elucidate the mode of action of TLN468 and its derivatives, a team-identified readthrough inducer that is more efficient than gentamicin and exhibits unusual selectivity regarding the incorporated amino acid at each stop codon.
Key contributions will include the design of quantitative reporters, implementation of functional screens, analysis of high-throughput data (NGS, ribosome profiling), and mechanistic interpretation of results.
Activities
- Molecular biology approaches (cloning, expression, in vitro transcription and translation).
- Biochemical approaches (protein purification, luciferase assays).
- Human cell culture, handling of lentiviruses.
- Development of a lentivirus-based screen for tRNA optimization.
- Presentation of work at team meetings and to collaborators.
