Job Description
As a researcher within the ANR EPIC project, you will investigate and optimise a cryo-PALM microscope and develop reliable cryogenic sample transfers between fluorescence and electron microscopy at IBS/IRIG. Key responsibilities include:
- Developing and evaluating optical improvements to the cryo-PALM microscope, focusing on excitation and emission paths, photon collection efficiency, spatial sampling, optical alignment, and quantitative point spread function (PSF) characterisation.
- Investigating and optimising illumination and acquisition strategies for cryo-FIB lamellae, addressing spatially targeted activation/excitation, sample heating control, and single-molecule localisation precision.
- Developing and validating protocols to transfer vitrified samples between cryo-FIB/SEM, cryo-PALM, and cryo-electron microscopy while maintaining cryogenic integrity and minimising contamination or deformation.
- Establishing calibration methods and assessing the overall performance of the correlative cryo-PALM/cryo-ET workflow alongside cryo-EM and image processing specialists.
- Contributing to the functional evaluation of fluorescent proteins utilised within the project.
Eligibility & Qualifications
Educational Qualifications
- A PhD in Optics, Photonics, Experimental Physics, Optical Instrumentation, or a closely related field.
Experience and Technical Skills
- Demonstrated hands-on experience in designing, aligning, optimising, and quantitatively characterising optical or microscopy setups.
- Knowledge of fluorescence microscopy, super-resolution imaging, cryogenic instrumentation, or biological applications is highly desirable.
- Prior experience with PALM or cryo-EM is welcome but not mandatory.
- Proficiency in programming and quantitative experimental data analysis is considered an asset.
Languages & Personal Competencies
- English: Fluent (written and spoken) for international research collaboration.
- French: Intermediate or basic knowledge is an asset, but not mandatory.
- Strong problem-solving mindset, scientific rigour, curiosity, and high autonomy.
- Ability to work within an interdisciplinary team combining optics, biophysics, and structural biology.
Benefits and Working Environment
- Access to world-class scientific infrastructure and cutting-edge biophysical facilities in Grenoble.
- Subsidised staff canteen/meal vouchers and coverage of up to 85% of public transport commuting costs.
- Active staff committee offering access to various cultural, sporting, and social activities.
- A scenic living and working environment nestled in the French Alps.
- CEA is committed to equality and inclusion; workplace accommodations are available for researchers with disabilities.
