Position 1:
About- NEUcrest is a four-year project, funded by the European Union Horizon 2020 Programme. The neural crest is an essential stem cell population of the vertebrate embryos. The project focuses on integrating academic, clinical and industrial research for a better understanding of neural crest development and neural crest related diseases. The NEUcrest network comprises 20 partners in academia, industry and hospitals from seven European countries. ESR5 – Modelling Neurocristopathies in Xenopus, mechanisms and drug screening ( supervised by Grant Wheeler (University of East Anglia, Norwich, UK)

Eligibility/Qualification–
1) First degree or Masters in Biological Sciences, Cell Biology, Genetics and Molecular Biology.
2) Mobility requirement: EU applicants are eligible to apply to all positions, non-EU applicants are eligible only to the position based in Israel.
Applicants must not have been based in the country of desired Ph.D. position for more than 12 months in the last 3 years prior to recruitment.
The standard minimum entry requirement is 2:1
Job/Position Description-
1) determine a temporal and spatial map of the development of the adrenal gland during Xenopus embryogenesis including how the NC contributes to its development.
2) generate novel assays for gene discovery and function of diseases associated with the adrenal gland. Potential syndromes to look at include Allgrove syndrome and Von Hippel-Lindau syndrome. Assays will include gene knockdown by morpholino or CRISPR/Cas9 and measurement of adrenaline and noradrenaline levels.
3) carry out a small molecule enhancer/suppressor screen on an adrenal gland Xenopus model line generated by the student.
The student will gain skills in molecular biology, embryology, in vivo biology and chemical biology. In addition, training for transferrable

MicroRNAs (miRNAs) are short, non-coding RNAs around 22 nucleotides in length that inhibit gene expression by translational repression or by causing degradation of target mRNAs. Some genes associated with miRNA production are deleted in neurocristopathies (e.g. DGCR8 in DiGeorge Syndrome).
Research group have identified two microRNAs (miR-196a and miR-219) expressed in the early neural crest. Our preliminary studies show that knockdown of either miRNA in the model organism Xenopus tropicalis (African clawed frog) causes a loss of neural crest tissue and are therefore predicted to target genes involved in NC specification.
In the ‘NEUcrest’ project, the student will:
1. identify potential targets of miR-196a and 219,
2. validate possible targets using cell-based luciferase assays
3. knockout target sites in specific genes by CRISPR/Cas9 and determine where miR-196a and 219 act in the gene regulatory network for NC development.
The student will gain skills in molecular biology, embryology, in vivo biology and chemical biology. In addition, training for transferable science skills in outreach and industrial management, are deeply embedded in the ITN programme.

ESR 13 project – Tissue environment and melanocyte differentiation: role of the adipocytes (supervised by Carmit Levy (Tel Aviv University, Israel)

– a Master degree in molecular biology awarded at the start of the Ph. D. position;
– Less than 4 years’ full time equivalent research experience and not yet been awarded a doctoral degree;

Job/Position Description- Â
The current Ph.D. research project aims at establishing the network of interaction between various neighbouring cells and melanocyte starting at early neural crest developmental stage, and to determine whether UVB radiation at any developmental stage will affect the cell-cell interaction and phenotype. It will also include investigation of whether an early developmental interaction between melanocyte and neighbouring cells may affect later melanoma formation. It will rely on generation of in-vivo and in-vitro systems for dissecting the molecular crosstalk between melanocyte and neighboring cells, including the focused on singling as Notch, WNT5a, WNT31 and TGFb, based on our published and unpublished knowledge. The genetic network controlling interactions between adipocyte/melanocyte will identify novel disease genes. Network understanding will identify novel therapeutic targets in prevention of melanoma formation starting at early developmental stages.
The successful candidate will be mentored by Prof. Carmit levy, a leading research in the melanoma field.
